Getting the Best Results from SAMi

Proper training and technique are essential for achieving optimal results with any CASA system. This page provides resources and guidance to help you get the most from your SAMi system.

Microscope Setup

Problems with image quality and especially microscopy will affect object recognition and therefore evaluation of every parameter. It is surprising just how many centers do not offer basic microscopy training and complain of poor image quality but simply have poorly adjusted phase contrast.

Images showing good contrast between background and white sperm heads (as shown above) are ideal for CASA analysis.

Sample Preparation Best Practices

The quality of your analysis begins with proper sample handling:

  • Allow samples to liquefy completely before analysis (typically 20-30 minutes)
  • Mix samples gently but thoroughly before taking aliquots
  • Allow samples to settle in the chamber before beginning analysis
  • Maintain temperature at 37°C for motility assessment where possible
  • Dilute samples exceeding 75-80 million/ml for optimal accuracy

Using the Edit Feature

The SAMi edit feature is crucial for maximizing detection accuracy. Before analysis, review the detected objects and:

  • Remove trackers from non-sperm cells (debris, leucocytes, germinal cells)
  • Adjust sensitivity settings for samples with unusual sperm morphology
  • Verify that all visible sperm have been detected

Understanding Sensitivity Settings

The variable sensitivity feature allows you to optimize detection for different sample types:

  • High sensitivity: Use for samples with small or faint sperm heads
  • Low sensitivity: Use for samples with significant debris to reduce false positives
  • Standard settings: Appropriate for most routine samples
